Extracts from Porcine Placenta Promote Proliferation of Mouse Embryonic Stem Cells

Embryonic stem (ES) cells are pluripotent cells derived from the inner cell mass (ICM) of blastocyst-stage embryos whose unlimited proliferative capacity has made them a potentially attractive source of pluripotent cells for various cell therapies. However, the full potential of ES cells will only be achieved when it becomes possible to expand them in high numbers without inducing spontaneous differentiation. Here we show that pig placenta extract has an effect on the growth of mouse ES cells and maintains ES cells in an undifferentiated and pluripotent state. Mouse ES cells cultured with placenta extract-treated media expressed alkaline phosphatase, and formed embryoid bodies (EBs) in vitro and teratomas in vivo. These results suggest that pig placenta extract may enhance the proliferation of ES cells without compromising their pluripotent properties.
