ADH Cluster Genes, Genome-Wide Association Studies, and Alcohol Dependence

Alcohol dependence (AD) is a multifactorial and polygenic disorder involving complex gene-to-gene and gene-to-environment interactions. Numerous studies have supported that genetic variations in a large number of genes affect a person's risk of AD. Among them, two major genes and their variants involved in alcohol metabolism-ADH1B and ALDH2-have distinct known effects on the risk of AD. Several genome-wide association studies (GWASs) and subsequent replication studies in Caucasian and African populations have identified other genes affecting the risk of AD and related traits. In addition, GWASs in East Asian populations have identified a number of genetic loci, including the ADH1B and ALDH2 on 12q24. Subsequent replication study has demonstrated that ADH1B His47Arg may be the sole functional genetic marker across the ADH gene cluster in East Asian populations. © 2016 Elsevier Inc.
